Isolated edited clones

• 2 “knocked-out” clones (both alleles) made with a single gRNA and 2 wild-type clones from the same pool of transfected cells that did not edit either allele;
• Maintenance of edited clones in our collection for six months, which can be resent for free (excluding post and package).
• Strategy design and testing of up to 6 gRNAs;
• Mycoplasma test at arrival and before shipment;
• Electroporation of RNPs and GFP-encoding plasmid;
• Sorting GFP+ cells;
• Confirmation by Sanger sequencing that the locus is being targeted by the Cas9 endonuclease;
• Isolation of clones;
• Genotyping of clones and confirmation of knockout by Sanger sequencing;
• Clone expansion, blind reconfirmation and shipment;

Edited bulk cultures

• 2 vials of “knocked-out” bulk cultures made with a single gRNA;
• Strategy design and testing of up to 6 gRNAs;
• Mycoplasma test at arrival and before shipment;
• Electroporation of RNPs and GFP-encoding plasmid;
• Sorting GFP+ cells;
• Confirmation by Sanger sequencing that the locus is being targeted by the Cas9 endonuclease;
• Maintenance of edited bulks in our collection for six months, which can be resent for free (excluding post and package).

“Do it yourself” package

Do it yourself” is a kit for 4 transfections, consisting of:

• Editing strategy;
• 3 gRNAs;
• Cas9;
• Tracer;
• Genotyping primers.
